About FOXO4-DRI
FOXO4-DRI was developed at Erasmus University Medical Center and published in Cell in 2017. The peptide uses a D-retro-inverso configuration for enhanced proteolytic stability. Research demonstrates that senescent cells rely on FOXO4-mediated sequestration of p53 to avoid apoptosis. FOXO4-DRI competes with endogenous FOXO4, releasing p53 to trigger mitochondrial apoptotic pathways selectively in senescent cells. Preclinical studies in aged mice showed restoration of fitness, fur density, and renal function. It represents one of the most targeted senolytic approaches in current research.
